If you are a reader of this Blog, you know I recently returned from The Mayo Clinic in Rochester MN, disappointed and dejected after having been denied entrance into their Clinical Stem Cell Trial for MSA. Shame on me for not having a back-up plan! After recovering from the pain of having been denied, I set out to find the next best set of trials available to MSA sufferers looking for a “Miracle”. This is part 3 of a 4 part series, in which I share the results of my month long research. Though I have ranked these for my own personal path forward, I will not share that with you until I start blogging about my participation in my #1 choice, sometime after January of next year. Part 3 of 4: “A Glimpse of Hope: My Perspective on the ATH434 Clinical Trial for MSA”

Living with Multiple System Atrophy (MSA) is a journey marked by challenges and the constant search for hope. The ATH434 clinical trial, sponsored by Alterity Therapeutics, represents a significant opportunity to realize that hope​​.

Understanding the ATH434 Trial:

  1. The Trial’s Scope: ATH434 is currently being assessed in a Phase 2 clinical trial, with 77 participants enrolled. The study follows a randomized, double-blind, placebo-controlled model, ensuring that the results are reliable and unbiased. This rigorous approach is vital in determining the true effectiveness of ATH434​​.
  2. Treatment and Comparison: Participants are divided into two groups, receiving either different doses of ATH434 or a placebo, taken twice daily. This comparison is crucial to understand how effective ATH434 is compared to not receiving the drug​​.
  3. Measuring Success: The primary outcome measure is a change in brain iron content, as iron accumulation is a key feature of MSA. Additionally, the trial looks at changes in aggregating alpha-synuclein levels, neurofilament light chain levels, the Unified MSA Rating Scale (UMSARS) score, and the SF-36 score. These varied measures provide a comprehensive view of ATH434’s impact​​.

Pros and Cons of the ATH434 Trial:

Pros:

  1. Focused Approach: By targeting specific biomarkers and symptoms of MSA, ATH434 could offer targeted relief.
  2. Comprehensive Assessment: The diverse outcome measures mean that any benefits of ATH434 will be thoroughly evaluated.
  3. Potential for New Treatment: If successful, ATH434 could become a vital part of managing MSA symptoms.

Cons:

  1. Limited Accessibility: The trial’s strict criteria, including age range and disease stage, mean not all MSA patients can participate​​.
  2. Uncertain Outcomes: Like any trial, there’s no guarantee of success or that ATH434 will be effective for all participants.
  3. Time to Results: With the trial estimated to complete in November 2024, patients and families must wait for conclusive results​​.

My Personal Reflection:

As someone grappling with MSA, the ATH434 trial symbolizes hope. It’s not just about a potential new medication; it’s about the ongoing fight against MSA and the search for better treatments. Participating in or following the progress of such trials gives us a sense of being part of something bigger — a community united in the fight against a common enemy.

Conclusion:

The ATH434 trial is a significant step forward in MSA research. For those of us living with this condition, every new trial, every bit of research, brings us closer to understanding MSA and potentially finding effective treatments. We watch this trial with hope and cautious optimism, knowing that each step forward, no matter how small, is a victory in our ongoing battle.

    Coach, my son is David Knox, also dealing with MSA. Stay hopeful and know there are many praying for some sort of positive outcome for all suffering with this terrible disease.
